Relationship between pluripotency and tumorogenesis

The potential formation of teratoma/teratocarcinoma from pluripotent stem cells represents one the main obstacles to safe use stem cell-based regenerative therapy. This study addresses the relationship between pluripotency and tumorogeneis using the colonial ascidian Botryllus schlosseri (Ascidiacea). This project takes advantage of an in vivo phenomenon (vascular budding, VB), where a population of pluripotent cells form a teratoma-like structure which eventually regain “correct” cell fate and positional information, in order to explore the molecular bases regulating the equilibrium between pluripotency and tumorigenicity in a chordate. Two complementary approaches are proposed: (1) transcriptome analyses of defined VB phases; (2) targeted study of endomesoderm specification during VB.
