Axes and “Germ layers” specification in non-embryonic

Wnt/beta-catenin is one of the earliest indicator for the establishment of the embryonic axis and formation of endomesoderm tissues in ascidians and other animals’ embryos. Herein, we first morphologically analyzed the area of initiation of the regenerative bud, prior to the observable establishment of axes polarity. We then analyzed the process of budding patterning by comparing the spatial-temporal expression of genes involved in the core (Axin, GSK3b), and downstream (Otx2, GSC, Bra, FoxA1, cMyc) canonical Wnt/beta-catenin pathway. The latter was used to identify the equivalent of the early germ layers specified domains in a non-embryonic scenario. Finally, we used a chemical approach and siRNA to infer the function and to analyze the modularity of the Wnt/beta-catenin pathway by studying the observed phenotypes and the modification of expression level of the downstream genes.
